Anti-Inflammation Activity of Naked Barley Betaone Prethanol Extract via Regulation of MAPK Pathway in RAW 264.7 Cells

Naked barley betaone is one of the newly developed barley varieties with higher ¥â-glucan contents. This study investigated the inhibitory effects of a naked barley betaone prethanol extract (BTE) on lipopolysaccharide (LPS)-induced inflammation in RAW 264.7 macrophage cells. BTE (100, 300, 500 ¥ìg/mL) dose-dependently decreased the production of nitric oxide (NO) and pro-inflammatory cytokines, such as tumor necrosis factor-¥á (TNF-¥á) and interleukin-6 (IL-6), in RAW 264.7 cells. The present study showed that the BTE treatment down-regulated inducible nitric oxide synthase (iNOS), cyclooxygenase 2 (COX2), TNF-¥á and IL-6 mRNA expression compared to the vehicle. Furthermore, the iNOS and COX2 protein levels were also down-regulated by the BTE treatment in LPS-stimulated RAW 264.7 cells. Finally, BTE suppressed the level of p38 and extracellular signal-regulated kinase (ERK) phosphorylation. Overall, BTE effectively inhibited LPS-induced NO production by suppressing the mitogen-activated protein kinase (MAPK) pathway in RAW 264.7 macrophage cells, which provided useful information for its application and development against inflammation.
